The duration of action of Botox is not determined by the potency of B-toxin...The toxin acts to destroy the synaptic connection and acts soon after the injection...And shortly after that most of the botox is gone...

Whats happens next is the the neuromuscular junctions continues to excrete neural growth factors which were the impetus for the nerve and the end organ to get together in the first place. So the duration of paralysis is determined by how long it takes the nerve to re-establish the connection back to the muscle...Not how long the drug remains potent...
