HPLC is just a final purification step, pump the bulk through a column and different components come out at different times. It's nothing to do with creating the bulk DNA sequences. The Vandalia patent is a continuous flow PCR for producing bulk DNA sequences. Typical PCR is done in fixed volume containers eg 96-well plates or small reaction tubes. The Vandalia method uses teflon tubing to allow a continuous flow of reactants.
