What a day so far, Amgen is at a all time high and reported a .61 dividend for next qrt. AMBS is set to take off and Sol is quite!
Since both of us are just guessing because neither one of us truely knows, I expect .25 by the end of March and 1.00 is possible! I'm accumulating is more accurate, started at .03 and have averaged up to .051. I have full confidence in AMBS's team and trust in their decisions!

Wolfram Syndrome is an extremely rare autosomal recessive disorder that affects only one out of 770,000 children. It causes insulin-dependent diabetes in childhood, as well as hearing and vision loss and kidney issues. As sufferers become older, they may also develop cognitive problems and dementia. Most die before they are 30. There is no treatment.
While scientists had previously identified brain abnormalities in sufferers from the disease, they had assumed that they came later on as the disease progressed. But a study led by Tamara Hershey, PhD, frin the Washington University in St. Louis, has found that these changes occur early in adulthood and even in young children with very mild symptoms.
Participants in the study received MRI scans and neurophysical testing, then were made to fill out a questionnaire. Control subjects – those with type 1 diabetes and those who were otherwise healthy – received just an MRI scan. They found that those with Wolfram Syndrome scored average or above average on tests for verbal intelligence, verbal learning, and memory.
For the brain scans, the study says, "We found reduced ICV [intracranial volume], dramatically reduced regional volumes and altered white matter microstructural integrity. The brainstem, cerebellum and optic radiations were particularly affected in WFS compared to both healthy controls and controls with type 1 diabetes. Notably, decreased brainstem and cerebellum volumes (after controlling for intracranial volume or ICV) were seen in almost all individuals with WFS, regardless of age or duration of diabetes or other features of the disease. These data together suggest that WFS affects brain development both globally and preferentially, even at the earliest stages of symptoms."
They also found differences in cortical thickness in areas of the brain, which are associated with vision, motor function, and higher-level function like memory. Sufferers from Wolfram Syndrome also were plagued with higher incidences of depression and anxiety.

Say the study authors, "Another possibility is that although there are early brain changes, these do not produce neurological symptoms until later in development, due to compensatory processes or the fact that the involved brain networks and functions do not come on-line until later."
The syndrome results from a mutation of a single gene, WSF-1, which was identified in 1998 by the late Dr. M. Alan Permutt. In 2010, he organized the world’s first clinic for the disease, testing and evaluating children with the disease. The third clinic will be held this summer.
Patient-based therapeutics part 6 - Wolfram syndrome iPSCs progress
I received many emails regarding our progress on Wolfram syndrome induced pluripotent stem cells (iPS cells) in the past two weeks. I would like to update you on a few things. As I mentioned in my previous blogs, we have created many iPS cells from skin cells of patients with Wolfram syndrome. These iPS cells can differentiate into various types of cells including brain cells and pancreatic beta cells that are damaged in patients with Wolfram syndrome
1. Disease modeling
We could successfully differentiate these iPS cells into neural progenitor cells. These are immature brain cells. We found that neural progenitor cells from patients are not completely damaged, which was surprising, but good news to us. Instead, they have altered calcium homeostasis. My impression right now is that cells from patients with Wolfram syndrome are "sensitive" to environmental stress, especially stimulus that changes cellular calcium levels. So we are looking for drugs that can modulate calcium homeostasis in cells to develop a treatment for Wolfram syndrome.
2. Testing drugs
As I mentioned above, we are focusing on drugs that can modulate calcium homeostasis in cells, especially endoplasmic reticulum calcium levels, to develop a treatment. Three drugs out of five candidate drugs that we have identified so far can control endoplasmic reticulum calcium levels. We are testing these three drugs using iPS cells.
3. Correcting a mutation
Using a special enzyme and artificial DNA, we are replacing an abnormal segment of Wolfram gene with a normal segment of Wolfram gene in patient-derived iPS cells. In theory, we should be able to correct altered calcium homeostasis through this process.
4. Making eye cells
A group in Columbia University Medical Center in New York could successfully make pancreatic beta cells from Wolfram syndrome iPS cells. We are collaborating with this group. So we are focusing our own efforts on making eye cells from Wolfram syndrome iPS cells. This is a collaboration project with a group in a major medical center in Japan. They have a special "recipe" for making eye cells. Because a clinical trial using this technology for an eye disease will start in a few weeks in Japan, I feel that this collaboration is so important for us. A physician and scientist who is working on this collaboration project will come to the US and work with us in a few months. The arrangement has been made, and the Japanese agency will partially support this effort.
