Chronic Anatabine Treatment Reduces Alzheimer's Disease (AD)-Like Pathology and Improves Socio-Behavioral Deficits in a Transgenic Mouse Model of AD.

Abstract
Anatabine is a minor tobacco alkaloid, which is also found in plants of the Solanaceae family and displays a chemical structure similarity with nicotine. We have shown previously that anatabine displays some anti-inflammatory properties and reduces microgliosis and tau phosphorylation in a pure mouse model of tauopathy. We therefore investigated the effects of a chronic oral treatment with anatabine in a transgenic mouse model (Tg PS1/APPswe) of Alzheimer's disease (AD) which displays pathological Aß deposits, neuroinflammation and behavioral deficits. In the elevated plus maze, Tg PS1/APPswe mice exhibited hyperactivity and disinhibition compared to wild-type mice. Six and a half months of chronic oral anatabine treatment, suppressed hyperactivity and disinhibition in Tg PS1/APPswe mice compared to Tg PS1/APPswe receiving regular drinking water. Tg PS1/APPswe mice also elicited profound social interaction and social memory deficits, which were both alleviated by the anatabine treatment. We found that anatabine reduces the activation of STAT3 and NF?B in the vicinity of Aß deposits in Tg PS1/APPswe mice resulting in a reduction of the expression of some of their target genes including Bace1, iNOS and Cox-2. In addition, a significant reduction in microgliosis and pathological deposition of Aß was observed in the brain of Tg PS1/APPswe mice treated with anatabine.
This is the first study to investigate the impact of chronic anatabine treatment on AD-like pathology and behavior in a transgenic mouse model of AD. Overall, our data show that anatabine reduces ß-amyloidosis, neuroinflammation and alleviates some behavioral deficits in Tg PS1/APPswe, supporting further exploration of anatabine as a possible disease modifying agent for the treatment of AD.

Why Axovant's $315M IPO bonanza should scare the hell out of you

Vivek Ramaswamy may not know anything about biotech and even less about treating Alzheimer's, but he just provided a master class on the current state of public investing and executing IPOs in the field.
The bare bones of Ramaswamy's new biotech venture, Axovant ($AXON), are much discussed these days. The former hedge fund manager grabbed an abandoned GlaxoSmithKline drug for Alzheimer's for $5 million. He gathered a team together and without recruiting a single patient for a pivotal study of a marginal drug designed to treat symptoms of the disease, just raised $315 million in an upsized IPO that came in at the top of the range and promptly gyrated much higher today as investors bought in.
Axovant, with no track record, no experience and one questionable product, leaped onto the market worth much more than $1 billion.
Given the fact that GlaxoSmithKline had a chance to take a look at this drug in the clinic, and concluded that they couldn't do better than selling it for lunch money, the IPO terms illustrate the kind of overnight riches a select few can find on Wall Street, provided you have the right kind of friends with money.
As Adam Feuerstein noted in a recent piece, two other hedge funds, RA Capital and Visium Asset Management, signaled an interest in buying up a bunch of these shares and were given a 90-day window to sell their shares, half the standard lock-up time.
None of this would be possible, of course, without the help of investors transfixed by the heady increase in the share prices of biotechs over the course of this bull market.
The fact that someone can make something of this size out of virtually nothing should be of concern to everyone in the industry. Magical thinking will take you just so far (remember the intoxicating dot-com days?). The argument that the market for biotech can stay stable rests on the belief that many of these new technologies in the clinic offer a fundamentally new and better way to treat many diseases. Something real. But when biotech mania takes over and perfectly legal schemes like this rain money, the pitfalls start to look like the Grand Canyon.

The 29 Year Old Behind The Giant Biotech IPO That Rose By 90% Speaks
This morning, a company that did not exist last summer had the biggest U.S. biotech IPO ever. Axovant Sciences sold shares at $15 a piece, raising $315 million at a $1.2 billion valuation. Already, the stock is up 90%, giving Axovant a valuation of about $2.8 billion.
Behind the massive IPO is a 29-year-old former hedge fund manager, Vivek Ramaswamy, who previously worked as a partner at Dan Gold’s QVT. In that capacity, we put him on the Forbes 30 Under 30 in Finance list. He left the hedge fund firm last year and founded Roivant, in which QVT is an investor. Axovant was formed in October and spun off by Roivant.
In December 2014, Ramaswamy engineered the purchase of an experimental Alzheimer’s pill from GlaxoSmithKline for $5 million. It is Axovant’s sole asset. GlaxoSmithKline also obtained a 12.5% royalty based on net sales and Axovant is obligated to pay the drug giant tens of millions of dollars more if certain milestones are achieved.

"Their New Zealand clinical trials appears (to me) headed in that direction. This trial may allow them to put on the label 'reduces chronic inflammation' legally. New Zealand is a British Commonwealth country so I would suspect it's laws are similar to England. I noticed they chose a Chinese doctor to run the trials. Is it possible that they may look to introduce the supplement version in New Zealand/Australia/Hong Kong and maybe followed by mainland China?"

Diana, your story matches mine, I am 55 and have had pancolitis for 25 years, it only goes into remission when I smoke, but, I hate smoking. The doctors said they wouldn't remove my colon unless I showed signs of cancer. I agree with everything you have written. I never knew the correlation between stopping smoking and getting a flare until the third or forth time it happened. My doctor did admit that they don't know what it was in the smoke that makes UC go into remission, but, just based on my experience, it isn't just nicotine since the gum and patches don't seem to work.

Phase 1 Clinical Studies to Begin Using Antabine Citrate as Possible Therapy for MS
Drug development company Rock Creek Pharmaceuticals, Inc. recently announced that a new clinical trial application with the United Kingdom’s Medicines Healthcare Products Regulatory Agency (MHRA) has been approved. The company is set to proceed with a Phase I study of its flagship pipeline product, Anatabine Citrate, a chemical that is found naturally in eggplant, potatoes, green tomatoes and other members of the Solanaceae family of plants, as well as in tobacco and tobacco smoke. The chemical is known for its anti-inflammatory properties unique from other anti-inflammatory drugs on the market, and may benefit patients with multiple sclerosis (MS).
The Phase I study will be composed of three portions, designed to evaluate Anatabine’s pharmacokinetic profile in the form of modified release formulation prototypes, and its safety and tolerability profiles in healthy volunteers. The first two parts will involve an open-label, non-controlled, single-dose study on 14 healthy participants, using six formula variations, with each administered dose spaced 7-14 days apart. The variations will be distinct in dose and duration of therapeutic action. This will allow the company to determine which formulation is most ideal, based on safety. The third and last part of the Phase I study will be a double-blind, placebo-controlled, seven-day multiple dose study of the identified optimal formulation in healthy subjects.
The Phase I clinical trial will base its success on two outcome measures, namely: “overall safety and tolerability through physical examination, vital signs, clinical chemistry, hematology, urinalysis and observation of adverse events (AE)” and “standard pharmacokinetics (PK) parameters and also pharmacodynamic (PD) assessment, specifically measuring the impact of the drug on inflammatory processes.”
“We are delighted to have been granted regulatory approval to begin our Phase I studies in the UK. This is the first clinical phase for our lead drug and will focus on safety and tolerability of six different formulations, 5 of which have modified release profiles and are of different doses. We look forward to generating our first human clinical data under this CTA,” said Dr. Michael Mullan, CEO of Rock Creek Pharmaceuticals.
Rock Creek’s UK-based partner, Quotient Clinical, is set to begin enrollment of healthy subjects this February 2015. While the company expects the study to stretch well into August, they expect to have a significant amount of research findings by mid-2015. The only surprise was you were running out of Anatabloc. As I recall you take 15 a day so that is 18.25 bottles a year. Fortuantely Ebay has had some attractive pricing--ie $ 75 to $90 bottle so it has not been hard to build up a supply over the last month.
I think we are going to see a time release version with much higher dosage of anatabine than we were taking with Anatabloc.

I don't believe it will be in Europe anytime soon. You can pick up bottles under $ 100 on ebay from time to time. I don't believe it will be available there for long. Perhaps a couple months. I am trying to extend my supply.

It is stated over and over (now) in the 10-Q that RCPI has PERMANENTLY EXITED the supplements market in the U.S. For example,
" On August 25, 2014, we received a response to the NDIN from the FDA. The letter indicated that the FDA considers anatabine, a principal ingredient in these products, to be a drug, because anatabine is intended to provide anti-inflammatory support, and was the subject of a previously filed INDA. Based on the FDA position, we permanently exited the dietary supplement business in the U.S. However, we will continue to seek opportunities to license the product for overseas markets."

It is amazing that you have to ask for the CRP test. My wife who had breast cancer a "year before Anatabloc-- 2011" has CRP of .03 now. In my mind at zero you stop aging. We monitor our CRP like hawks.
In order for her to get breast cancer her CRP had to go through the roof.
