In my opinion, PFS is a bogus “endpoint” that doesn't tell you squat.

Example 1:

Patient “A” has a PR with a 90% reduction in tumor volume down to 10 mm. After six months of treatment the tumor volume increases to 12 mm and the patient qualifies as having a progressive disease.

Patient “B” has a PR with a 30% reduction in tumor volume down to 70 mm. After six months of treatment the tumor volume increases to 84 mm and the patient qualifies as having a progressive disease.

Example 2:

Cohort “A” has a 64% OR rate and a 50% reduction in median tumor volume that increases to 60% of the original volume at 6.5 months.

Cohort “B” has a 35% OR rate and a 0% reduction in median tumor volume that increases an additional 20% at 6.2 months.
