I do not think that S1 agonism is the ONLY aspect that gives 2-73 its mechanism of action. I frankly do not think the mechanism of action at all is well understood. What is known - S1 and M1 receptors are manipulated by 2-73. There are likely other targets too - not yet identified.

I like to use the example of Depakote - a long known antiepileptic medication however we do not know its exact mechanism of action and why it works as an antiepileptic agent. Some related it to NMDA receptor effects. IT is likely multimodal.

So I'm not as caught up in knowing the exact MOA of 2-73 and getting burdened by the contradictions that donepezil is a stronger S1R agonist than 2-73 and donepezil doesn't work for cognitive stabilization.

IT would be interesting to know further how the cannabinoid system is involved in regulating neuronal homeostasis and I think there have been several articles pointing to how 2-73 plays a role in the endo-cannabinoid system - of which we are just scratching the surface in our understanding. Watch to see more mention of the endo-cannabinoid system and its role in "Cellular homeostasis" theory that Anavex is actively promoting.

Tip of the iceberg is right - we are only beginning to understand but the more data we get, the more questions seem to arise? The mystery unravels just as it gets more complicated, it would seem.
