350 mg of Prurisol was proven to be bioequivalent to 300 mg of Ziagen in Phase 1. The bioavailability is 25% for P and 83% for Z, meaning that 87.5 mg of P and 249 mg of Z will reach the systemic circulation.

If 249 mg of P needs to reach the systemic circulation, one would need to take almost 1000 mg of P. Here's my calculations.

249 mg (Z) / 87.5 mg (P) = 2.85

350 mg (P) * 2.85 = 997.5 mg

997.5 mg (P) * 0.25 (bioavailability) = 249 mg (P)

200 mg of P was shown to be effective in Phase 2a. Even at 400 mg, P is still half of Z in bioavailability.
