There are several papers talking about p21 mRNA qPCR results in cancer cell lines, such as this one for a sirtuin (HDAC class III) inhibitor:

p21 increases 5-8 fold, in a dose dependent fashion.

Again, as I mentioned in the above post, p21 increase may not always be desired, and cancer biology is very complex.

"Therefore, p53-induced apoptosis can be blunted by p21, and removal of p21 can enhance p53-induced cell death."

Here is a paper combining Nutlin-3a (to activate p53) with an XIAP inhibitor drug showed p21 *reduction* improved apoptosis in AML cell lines.

I'm not sure about clinical results. I may have seen some p53 activating drugs with early results, including p21, but I can't find them easily if there are any.
