With -L, the DCs are spoon fed the antigens. The tumor cells are exploded so that the lysate is all mixed in with the maturing DCs.
With Direct, the cells are partially matured and then injected into the tumor. They will have to hunt out antigens to recognize within the tumors home environment that is often known to be immuno suppressive.
A kid being spoon fed to eat does not mean he can go into the woods and hunt for game to live on.
There have been plenty of DCs variants trials. And many have failed. One success does not mean they all work.
