I believe that Dr. Coates is no longer being retained by CDEx as a PR prop.
I believe that after the failures of the Valimed fiasco at the ASHP Show, Dr. Stevenson has distanced himself from the embarrassment of CDEx.
Their new Spam Man (TM- d4diddy) now appears to be Dr. Coates.

If one reviews Vikings posts you will see my point. Research projects performed by Dr. Coates et al for other companies which are patented and owned by those companies have nothing to do with CDEx.
It is a deceptive way of exploiting the personal accomplishments of those individuals.

Then ask him when we can expect an actual working model {not an OTS RS prop} that performs all the functions as advertised.

The key phrases: sampling apparatus and optical structure
In the case the old Valimeds, the sampling apparatus was a cuvette which allowed for consistent and reliable measurement of concentration (but still only 10% accurate which cannot be considered an analytical instruments by any stretch of the imagination). The cuvette had been allegedly designed (and patented) to have a highly precise wall thickness or optical structure so all measurements would be consistent.
Trying to measure concentration through standard bottles, IV bags... etc. would require a different signature for every different type of container, even if the drug/dilutant inside were identical in each case. Unlike with the use of a cuvette, the great variations in how the sampling light reacts with the different containers makes the use of a single signature for a specific drug/dilutant (regardless of the container) impossible. A different signature would be required for each different container.
By trying to make it simpler to use they've increased the complexity in another way. It's outta the fire and into the frying pan so to speak.
So That is only one of the problems with the alleged claim that the iG4 can accurately measuring concentration without the use of a cuvette. Even for a given container (e.g. an IV bag) the variation in each bags wall thickness makes accurate measurement of concentration difficult (if not impossible) to be viable. That's why they had to design the special cuvettes in the first place.

No, the "how much" or concentration of a material present is directly proportional to the intensity of the return signal (light) at a given wavelength (Beer-Lambert Law- first order approximation).
As for the second part of your question, I have no idea what you're even trying to ask, but the optical signature consists of a unique pattern of absorptiom peaks at various wavelengths.
Intensity -> concentration
Absorption peak pattern -> signature
I believe the reason why CDEx can't get the G4 to measure drug concentration accurately is because they're trying to eliminate the hassel of using a cuvette and they can't get it to work without one. There is too much variation in the wall thickness of the vials, IV bags, syringes... etc. to get an accurate concentration reading.

Purchasing a Raman spectrometer to identify a few drugs for staged demos is most likely as far as the CDEx scamsters every got. Measuring the molar concentration of drugs (esp. multi-constituent drugs) through their containers was an absolute pipe dream.
An attractive concept to sucker investors out of their money, but just not possible.

Pursuant to each Line of Credit Agreement, the Company has issued each lender a Revolving Note (collectively, the “Notes”) evidencing the net amount drawn by the Company during term of the Line of Credit Agreement. The Notes bear interest at a rate of 12% per annum. Interest on the balance of the Notes accrues and is payable on the first day of each month beginning on November 1, 2014. On the maturity date, the Company must pay to the lenders an amount equal to 110% of the principal due on the Notes plus any accrued and unpaid interest as of that date.
