AUSTIN, Texas, May 30, 2023 (GLOBE NEWSWIRE) -- Plus Therapeutics, Inc. (Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced that a new analysis from its partner Medidata, a Dassault Systèmes company, was accepted for online publication at the 2023 American Society of Clinical Oncology (ASCO) Annual Meeting, taking place June 2-6, in Chicago, Illinois.
The abstract titled, “Clinical characterization of patients with recurrent glioblastoma in trials involving CED and non-CED treatment [#417610],” included data sourced from Medidata’s more than 30,000 clinical trials involving more than nine million de-identified participants. Aggregate summary statistics comparing bevacizumab and convection-enhanced delivery (CED) patients were based on combined study-level and patient-level data using sample size weighted means, incidence and overall survival.
A total of 799 patients were evaluated from MEDS and studies referenced in D’Amico [J Neurooncol 2021]. Both cohorts (bevacizumab and CED, respectively) were comparable in terms of median age (56 years vs. 55 years), Caucasian race (91% vs. 93%), male sex (68% vs. 65%) and median overall survival (7.9 months vs. 8.4 months).
“The purpose of this analysis was to determine whether bevacizumab patients could form an appropriate external control for evaluating overall survival in current and upcoming CED trials,” said Norman LaFrance, M.D., Chief Medical Officer of Plus Therapeutics. “The results of this analysis provide confidence in the use of an external control arm formed from aggregated clinical trial data of patients previously treated with bevacizumab to interpret the results of the ReSPECT-GBM Phase 1/2 trials. Thus far, our clinical outcomes are promising compared to those using an external control. In a potential future pivotal trial, the use of a historical control arm will enable Plus Therapeutics to more rapidly enroll the trial while simultaneously reducing trial costs.”
In April 2022, the Company entered into an expanded partnership with Medidata to utilize the Synthetic Control Arm® platform in its Phase 2 trial for rGBM, in a manner that has historically been favorably received by the U.S. Food and Drug Administration.

Medidata’s Synthetic Control Arm (SCA) – a type of external control – is formed by carefully selecting patients from Medidata’s extensive repository of historical clinical trials to match the baseline demographic and disease characteristics of the patients treated with the new investigational product. Case studies have shown that SCAs can effectively mimic a classic randomized control and, therefore, can be used to accurately interpret the treatment effects of an investigational product.
SCAs can help enhance the scientific validity of single-arm trials and, in certain indications, enhance randomized clinical trials to expose fewer prospective patients to control and/or ineffective or existing standard-of-care treatments that might not provide a benefit to the patient. The new supply agreement builds on the Master Services Agreement which the Company and PPS entered into in 2021 for the development, manufacture and supply of rhenium (186Re) obisbemeda.

(Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced the completion of enrollment of the original three patients required in Cohort 8 of the ReSPECT-GBM Phase 1/2a dose escalation clinical trial of rhenium (186Re) obisbemeda for the treatment of recurrent glioblastoma (GBM). Within the dose escalation phase of the trial, the Company has treated 27 patients to date.
In Cohort 8 of the Phase 1/2a trial, patients with large-sized tumors were administered a 41.5 mCi dose of rhenium (186Re) obisbemeda within an infused volume of 16.34 mL. In parallel, the Company is advancing a Phase 2b clinical trial evaluating rhenium (186Re) obisbemeda in recurrent GBM with small- to medium-sized tumors and also has a retreatment option for qualifying patients who have already received rhenium (186Re) obisbemeda.

Plus Therapeutics, Inc. (Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced the completion of Cohort 3 of the ReSPECT-LM Phase 1/2a dose-escalation clinical trial of rhenium (186Re) obisbemeda for the treatment of leptomeningeal metastases (LM) from solid tumors less than one month since its initiation, demonstrating rapid enrollment. Additionally, the Company has expanded the number of clinical trial sites to include Northwestern Memorial Hospital in Chicago, marking the first expansion of the ReSPECT-LM trial beyond Texas. Thus far, 10 patients have been treated across three cohorts in the ReSPECT-LM Phase 1/Part A dose escalation clinical trial.

Previous Phase 1 data presented at the Society for Neuro-Oncology Annual Meeting in November 2022 has shown that a single administered dose of rhenium (186Re) obisbemeda was feasible, safe, and well-tolerated. (Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced the first patient in Cohort eight of the ReSPECT-GBM Phase 1/2a dose escalation clinical trial evaluating the Company’s lead investigational targeted radiotherapeutic, rhenium (186Re) obisbemeda, in recurrent glioblastoma (GBM) has been successfully treated.
The dose escalation phase of the trial has now treated 25 patients with no dose-limiting toxicities observed through 7 completed dosing cohorts. Cohort 8 is now enrolling using 41.5 millicuries (mCi) administered dose rhenium (186Re) obisbemeda with an infused volume of 16.34 milliliters (mL) which allows the treatment of large and morphologically complex tumors.
In parallel, the Company has treated five patients to date with rhenium (186Re) obisbemeda in the Phase 2 trial in recurrent GBM administering a dose of 22.3 mCi and 8.8 mL in small- to medium-sized tumors.

Thus far, no treatment emergent dose-limiting toxicities have been observed in either trial. The absorbed radiation doses from the 30 patients treated to date have resulted in an absorbed dose of up to 740 gray to the tumor.

This follows successful prior completion of Cohorts 1 and 2 as well as the recommendation of the trial’s Data and Safety Monitoring Board (DSMB) to advance into Cohort 3. Thus far, no dose-limiting toxicities have been observed with administered radiation doses of up to 26.4 millicuries. This dose represents a theoretical maximum absorbed radiation dose to the cerebral spinal fluid of approximately 200 gray.

At the 2022 Annual Scientific Meeting and Education Day of the Society for Neuro-Oncology (SNO), Plus Therapeutics presented Phase 1 data from the ReSPECT-LM trial demonstrating that a single administered dose of rhenium (186Re) obisbemeda was feasible, safe, and well-tolerated. The ReSPECT-LM trial is a multicenter, sequential cohort, open-label, single dose, dose escalation Phase 1/2a study using a modified Fibonacci 3+3 study design. It will evaluate the maximum tolerated dose, maximum feasible dose, safety and efficacy of a single administration of rhenium (186Re) obisbemeda via intraventricular catheter for LM following standard surgical, radiation and/or chemotherapy treatment. The primary endpoints of the study are the incidence and severity of adverse events/serious adverse events and dose limiting toxicities. Secondary endpoints include overall response rate, duration of response, progression free survival and overall survival. (Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced completion of enrollment in Cohort 2 of the ReSPECT-LM Phase 1/2a dose escalation clinical trial of rhenium (186Re) obisbemeda for the treatment of leptomeningeal metastases (LM).

At the 2022 Annual Scientific Meeting and Education Day of the Society for Neuro-Oncology (SNO), Plus Therapeutics presented Phase 1 data from the ReSPECT-LM trial demonstrating that a single administered dose of rhenium (186Re) obisbemeda was feasible, safe and well-tolerated across two dosages in four patients from Cohorts 1 and 2, with patients experiencing a decreased cerebrospinal fluid tumor cell count at 48 hours following treatment of 46% to 92%.
The ReSPECT-LM trial (NCT05034497) is a multicenter, sequential cohort, open-label, single dose, dose escalation Phase 1/2a study using a modified Fibonacci 3+3 study design. It will evaluate the maximum tolerated dose, maximum feasible dose, safety and efficacy of a single administration of rhenium (186Re) obisbemeda via intraventricular catheter for LM following standard surgical, radiation and/or chemotherapy treatment. The primary endpoints of the study are the incidence and severity of adverse events/serious adverse events and dose limiting toxicities. Secondary endpoints include overall response rate, duration of response, progression free survival and overall survival.
The U.S. Food and Drug Administration has granted Fast Track designation to rhenium (186Re) obisbemeda for the treatment of LM. ReSPECT-LM is funded by a three-year $17.6 million grant by the Cancer Prevention & Research Institute of Texas (CPRIT).
About Leptomeningeal Metastases (LM)
LM is a rare complication of cancer in which the disease spreads to the cerebrospinal fluid (CSF) and central nervous system that can originate from solid tumors, primary brain tumors, or hematological malignancies. Breast cancer is the most common cancer linked to LM, with 3-5% of breast cancer patients developing LM. Lung cancer and melanoma can also spread to the CSF and result in LM. LM occurs in approximately 5% of people with cancer and is usually terminal with one-year and two-year survival of just 7% and 3% respectively. The incidence of LM is on the rise, partly because cancer patients are living longer and partly because many standard chemotherapies cannot reach sufficient concentrations in the spinal fluid to kill the tumor cells, yet there are no FDA-approved therapies specifically for LM.

AUSTIN, Texas, Jan. 18, 2023 (GLOBE NEWSWIRE) -- Plus Therapeutics, Inc. (Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing targeted radiotherapeutics with advanced platform technologies for central nervous system cancers, today announced that the first patient has been dosed in the ReSPECT-GBM Phase 2b dose expansion clinical trial evaluating rhenium (186Re) obisbemeda for the treatment of recurrent glioblastoma (GBM).
This Phase 2b multi-center trial is designed to evaluate the safety, tolerability, distribution and efficacy of rhenium (186Re) obisbemeda infused directly into the tumor via convection-enhanced delivery catheters in patients with recurrent GBM progressing after conventional treatment.
“In the Phase 1/2a dose escalation trial, we showed that a rhenium (186Re) obisbemeda radiation dose of 22.3 mCi in an infused volume of 8.8 mL can be safely administered and that there is a statistically significant correlation between overall survival and both absorbed radiation dose to the tumor and percent tumor volume in the treated volume,” said Andrew J. Brenner, M.D., Ph.D., Professor of Medicine, Neurology, and Neurosurgery, Kolitz/Zachry Endowed Chair Neuro-Oncology Research, and Co-Leader of the Experimental and Developmental Therapeutics Program at The University of Texas Health Science Center at San Antonio and principal investigator of the ReSPECT-GBM clinical trial. “The strength of this correlation is unusually positive for a Phase 1/2a trial and we are optimistic that these safety and efficacy signals will be confirmed in the ongoing Phase 2b trial.”
The Phase 2b trial is expected to enroll up to 31 additional patients with small- to medium-sized tumors (20 mL or less) in approximately 24 months. The trial is supported by an award from the National Cancer Institute (NCI), part of the U.S. National Institutes of Health (NIH).
“We have successfully completed two of our key near-term clinical development goals, specifically to manufacture cGMP rhenium (186Re) obisbemeda and move our glioblastoma program into Phase 2b,” said Norman LaFrance, M.D., Chief Medical Officer and Senior Vice President at Plus Therapeutics. “The treatment of our first patient went well with excellent targeted tumor delivery of high dose rhenium (186Re) obisbemeda, similar to that seen at this same dose in our Phase 1/2a trial. As disclosed at the Annual Scientific Meeting and Education Day of the Society for Neuro-Oncology in November 2022, results from the Phase 1 ReSPECT-GBM clinical trial of rhenium (186Re) obisbemeda in 24 patients with recurrent GBM demonstrated that a statistically significant improvement in overall survival correlated with both the average absorbed dose of radiation to the tumor and the percent volume of tumor treated. The treatment was safe and well tolerated without dose limiting toxicities.

About Recurrent Glioblastoma (GBM)
GBM affects approximately 14,490 patients annually in the U.S. and is the most common and lethal form of brain cancer. The average life expectancy with GBM is less than 24 months, with a one-year survival rate of 40.8% and a five-year survival rate of only 6.9%. There is no clear standard of care for recurrent GBM and even the few currently approved treatments provide only marginal survival benefit and are associated with significant side effects, which limit dosing and prolonged use. Approximately 90% of patients experience GBM tumor recurrence at or near the original tumor location, yet there are no FDA-approved treatments in the recurrent or progressive setting that can significantly extend a patient’s life.

(Nasdaq: PSTV) (the “Company”), a clinical-stage pharmaceutical company developing innovative, targeted radiotherapeutics for rare and difficult-to-treat cancers, today announced that the World Health Organization’s (WHO) International Non-proprietary Name (INN) Expert Committee has selected the non-proprietary name “Rhenium (186Re) obisbemeda” for the Company’s lead investigational targeted radiotherapeutic, formerly known as 186RNL. Going forward, Plus Therapeutics will use Rhenium (186Re) obisbemeda in place of 186RNL.

Dr. Brenner holds the Kolitz/Zachry Endowed Chair Neuro-Oncology Research and is the Co-Leader, Experimental and Developmental Therapeutics Program at UT Health Science Center San Antonio (UTHSCSA) as well as a practicing oncologist at Texas Oncology in Austin. His experience with first-in-human studies includes involvement with the development of approximately 30 compounds as either a co-Investigator or Principal Investigator since 2008. This also includes first-in-class experience (gene therapy, liposomal encapsulated microRNA mimic, liposomal encapsulated therapeutics, metabolic inhibitors, etc.). He is familiar with all phases of study conduct.
Dr. Brenner is a graduate of Texas A&M University where he earned his bachelor’s degree in biochemistry and went on to earn his doctorate in biological science and tumor biology at The University of Texas M.D. Anderson Cancer Center Science Park.
