The reason the first version didn't work was because of the "chaff," the soluble / secreted GP protein. EbolaCide1 was designed to go after that part of the virus, but the virus sheds that particular protein to throw off the immune system. Thus EbolaCide1 attacked the chaff and not the virus, much like the immune system being tricked into going after the wrong protein.

They thought that designing EbolaCide2 to go after the part of Ebola that binds to NPC1 would work better, but that part is capped until the very end, and EbolaCide2 couldn't find that part of the virus to attack it well either.
